REPRODUCTIVE RESPONSE OF RABBITS FED SUPPLEMENTAL Moringa oleifera (Lam) LEAF MEAL

- BACKGROUND: Oral melanin hyperpigmentation (OMH) has been classified as a HIV associated condition which may present as a brown-black macule or patch of the oral mucosa in HIV seropositive patients. HIV-OMH may be idiopathic, drug- induced or due to adrenal insufficiency. This cross-sectional study was conducted to determine the prevalence of HIV- OMH among HIV seropositive patients attending the Infectious Disease Institute centre, UCH Ibadan. METHODOLOGY: Consecutive, consenting HIV seropositive patients attending PEPFAR clinic, UCH, Ibadan were enrolled. Data collected included age, gender, duration since diagnosis of HIV and commencement of HAART, history of smoking, any systemic disease, WHO staging of HIV, CD4 count, presence of OMH and site affected. Data analysis was done using SPSS version 15. RESULTS: The study group of 150 HIV sero-positive patients comprised 24 males (16%) and 126 females (84%). Out of all the patients seen, OMH was seen in 97 of them, majority reported not being aware of the condition, some noted it before being diagnosed of HIV, while only 14(14.4%) reported the presence of OMH after being diagnosed with HIV and commencement of HAART (p= 0.032). Concerning the OMH sites, gingiva was most noted followed by tongue, palate, buccal and labial mucosa. CONCLUSION: The prevalence of HIV-OMH in this study was 14.4%. Gingiva was the most common site affected. 2 results 2

- Abiotic stress such as heavy metal contamination of agricultural soil limits crop production by disrupting plant’s physiological activities and seed germination. Exogenous proline, hormone and compost have been used to induce growth and stress tolerance in crop but little is known about their interactive and anti-oxidative roles in crop exposed to heavy metal stress. This study assessed the effects of pre-sowing seed soaking with exogenous proline (P) and hormones (Gibberellic acid; GA and Indole-acetic acid; IAA) on the Pb accumulation, osmolytes (P, phenolics; PH and Glycine betaine; GB), Photosynthetic pigments (chlorophyll and carotenoids) and stress indexes (H2O2 and malonialdehyde; MDA) of maize grown on compost amended Pb (23,959 mg kg-1 Pb) contaminated soil. Compost was applied at 20 t ha-1 and 40 t ha-1, P, GA and IAA were applied at 10 mM and 5 mM solely and in combinations to give a total of 34 treatments including control (Distilled water only) and replicated 3 times. Except in IAA treatment, seed pre-treatments with exogenous P and GA and soil amendment with compost generally enhanced chlorophyll production in maize and reduced GB, carotenoids, H2O2 and MDA in treated seeds compared to control. With regards to stress indexes G2+C1 gave the lowest values. Sole application of P1, P2, C2, G1 and I2 enhanced the production of proline in treated plants compared to combined application. Combination with compost was better than hormonal combination. A combination of P2 with C (40 t ha-1) (P2C2) gave the maximum chlorophyll content and reduced the endogenous GB and P. Treatment with P2C2 also reduced Pb accumulation in plant. In conclusion, exogenous application of hormones and proline in combination with organic amendment induces heavy metal tolerance in maize and increased protection against oxidative stress compared to contaminated control. The technique that can enhance crop growth on contaminated soil, reduce metal uptake and provide osmo-protection could be a promising approach. 1 results 1
- Africa has great intensity of chemical exposure and high level of infertility. Functional disorders of the male germ cell and endocrinopathies have been attributed to exposure to mixed chemicals. Data on the mixed chemical exposure on reproductive hormones are sparse in Nigeria. This study was designed to evaluate male reproductive hormones and determinants of occupational exposure to mixed chemicals in a mechanic village in Ibadan. Forty-three males, auto mechanics aged 18-60 years occupationally exposed to mixed chemicals (MCG) at their work place in Bodija mechanic village with mean±SEM duration of 21.2±1.9 years were age matched with 40 unexposed males (controls) from the University College Hospital, Ibadan and environs. Demography, anthropometry, blood pressure and sexual history were obtained by standard methods. Blood (10 ml) was obtained from each participant for estimation of reproductive hormones by enzyme linked immunosorbent assay while total antioxidant capacity (TAC) was determined spectrophotometrically. Testosterone/oestradiol ratio (TE ratio) was calculated. The MCG were classified based on their reproductive hormone levels into 4 subgroups as eugonadic, hypogonadotrophic hypogonadism, sub-optimal hypogonadism and compensated hypogonadism. P<0.05 was regarded as significant. There were significantly raised oestradiol levels, diastolic blood pressure, waist circumference and waist to hip ratio and significantly lower TAC and TE ratio in the MCG than the controls (P<0.01). Exposed hypogonadic males exhibited significantly lower LH, FSH, testosterone, prolactin, TAC levels and TE ratio than exposed eugonadic males (P<0.05). Hypogonadism in the males exposed to mixed chemicals may be associated with reduced TE ratio resulting from increased adipose mass as well as oxidative stress associated with the mixed chemical exposure. 1 results 1

- Background Endometrial carcinoma is the commonest genital tract malignancy in most developed nations, but it lags behind cervical carcinoma and ovarian cancers in most developing nations, including Nigeria. Estrogen has been described as a promoter of endometrial carcinogenesis. Objectives The aim of this study was to demonstrate the frequency of estrogen receptor (ER) and progesterone receptor (PR) expressions in endometrial carcinoma and to correlate them with tumour grade. Materials and Methods Cases of endometrial carcinoma diagnosed in the Department of Pathology over a 10-year period were reviewed retrospectively. The paraffin-embedded blocks were retrieved, and immunohistochemistry for ER and PR was performed on them. Haematoxylin and eosin (H&E) slides were reviewed, and tumours were graded by three independent pathologists. Data were analysed using SPSS version 22. The level of significance was set at P ≤ 0.05. Results There were 44 cases of endometrial carcinoma. ER and PR were positive in 29.5% and 18.2% of cases, respectively. There was no significant association between ER (P = 0.361) and PR (P = 0.204) expressions and histological grade of the tumour. The most common histological grade was grade 3, with 70% of cases (36 cases), whereas 13 cases (26%) were grade 2 and only 2 cases (4%) were grade 1. Conclusion The positive expressions of ER and PR in endometrial carcinoma suggest that steroid receptor studies may be of potential benefit in the management of some patients with endometrial carcinoma. Future studies employing larger sample sizes are therefore recommended. 1 results 1

- Background: Genital Chlamydia infection (GCI) and the associated pathologies have been implicated in tubal infertility. Though the actual pathologic mechanisms are still uncertain, oxidative stress and other factors have been implicated. The purpose of the study was to determine the possible contribution of female reproductive hormones and biomarkers of oxidative stress in genital Chlamydial infection to tubal occlusion. Methods: This prospective case control study was carried out by recruiting 150 age matched women grouped into infertile Chlamydia positive women (n=50), fertile Chlamydia positive women (n=50) and fertile Chlamydia negative women as controls (n=50). High vaginal swabs and endocervical swabs were collected for screening Neisseria gonorrhoeae, Chlamydia trachomatis, Trichomonas vaginalis, Treponema pallidum, Staphylococcus aureus, and Candida albicans. Sera were collected for estimation of Chlamydia trachomatis antibody, female reproductive hormones [Follicle Stimulating Hormone (FSH), Luteinizing Hormone (LH), Oestradiol (E2), Progesterone (P4), Prolactin (PRL)] and biomarkers of oxidative stress [Total Antioxidant Capacity (TAC) and 8-hydroxyl-2-deoxyguanosine (8-OHdG)] by enzyme immunoassay (EIA). Data were analyzed using chi square, analysis of variance and LSD Post hoc to determine mean differences at p=0.05. Results: Among women with GCI, higher levels of LH and 8-OHdG were observed in infertile Chlamydia positive women compared to fertile Chlamydia positive women (p<0.05). Higher levels of LH and 8-OHdG and lower TAC levels were observed in infertile Chlamydia positive women compared to fertile Chlamydia negative controls (p<0.05). Conclusion: Mechanisms including oxidative DNA damage and reduced antioxidant capacity may be involved in the pathology of Chlamydia induced tubal damage. 1 results 1
- Background: Pituitary and gonadal dysfunctions resulting from increased adiposity leading to disturbances of sexual and reproductive functions have been reported in males with metabolic syndrome (MS) and type 2 diabetes mellitus (DM2). The aim of this study was to evaluate sexual dysfunction, leptin, and reproductive hormones in Nigerian males with MS and DM2. Methods: Participants were 104 men (34 males with DM2, 17 men with MS and 53 men with normal body mass index (18.5-24.9 Kg/m2) without MS (controls)). The International Diabetes Federation (2005) criteria were used for MS diagnosis. Reproductive history, anthropometry, blood pressure (BP) and 10 ml fasting blood samples were obtained by standard methods. Fasting plasma glucose, total cholesterol, triglycerides and high density lipoprotein cholesterol were determined by enzymatic methods while low density lipoprotein cholesterol was calculated. Leptin, follicle stimulating hormone (FSH), luteinising hormone (LH), prolactin, testosterone and oestrogen were determined by enzyme immunoassay (leptin by Diagnostic Automation, Inc.; others by Immunometrics (UK) Ltd.) while oestrogen-testosterone ratio was calculated. Data analyzed using ANOVA, Chi square and multiple regression were statistically significant at p<0.05. Results: Testosterone was significantly lower in MS than controls while oestradiol and ETR were significantly higher in MS compared with controls and DM2 group (p<0.05). ETR significantly predicted testosterone in all groups (p<0.05). Significantly lower libido was observed in men in MS than controls and DM2 groups (p<0.05). Conclusion: Sexual and reproductive dysfunction may be related to increased conversion of testosterone to oestrogen in increased adipose mass in men with metabolic syndrome and type 2 diabetes mellitus. 1 results 1
