- Adequate information on how leachates affect hematological and reproductive functions is necessary to help in linking causality with predictable response. The present study investigated the effects of Olushosun municipal landfill leachate (OMLL) exposure and withdrawal on sperm characteristics and erythrocytes oxidant–antioxidant balance in rats. Adult male Wistar rats were exposed to 0%, 12.5% and 25% OMLL in drinking water for 28 days. One half of the rats in each group were sacrificed on day 29 while the remaining one-half stayed an additional 28 days without treatment. OMLL exposure significantly decreased sperm functional parameters, disrupted antioxidant systems with concomitant elevation in hydrogen peroxide and malondial dehyde levels in erythrocytes and sperm. Following withdrawal of treatment, OMLL-mediated decrease in sperm count and daily sperm production were reversed to near control. However, erythrocytes and sperm oxidative damage, increased sperm abnormalities, decreased epididymis weight, sperm progressive motility and testicular sperm number persisted and were consistent with results obtained from rats sacrificed immediately after OMLL treatment. Collectively, OMLL-induced irreversible oxidative damage to erythrocytes and sperm in rats within the time course of investigation. These findings highlight potential adverse effects of OMLL on individuals unduly exposed to leachates contaminated substances. 1 results 1

- Cobalt stimulates erythrocyte production via mechanisms that mimic physiological adaptations to hypoxic conditions. However, little is known about alterations in the balance of erythrocyte antioxidant defense system produced by cobalt. We investigated the effect of Kolaviron (KV) on cobalt chloride (CoCl2)-induced disturbances in erythrocyte antioxidant status and hematological parameters and compared the effects with those of Gallic acid (GA). Groups of rats were orally treated with either KV1 (100 mg/kg), KV2 (200 mg/kg) or GA (120 mg/kg), along with CoCl2 (350 ppm) in drinking water for 14 days. CoCl, produced significant (p<0.05) increases in packed cell volume, hemoglobin and red blood cell count, but no alterations in erythrocyte morphology, in the same way as rats treated with KV or GA. Significant (p<0.05) elevation in malondialdehyde (MDA) content and reductions in total thiols and reduced glutathione (GSH) in the CoCl2 group were indications of oxidative stress. KV produced significant (p<0.05) reduction in MDA, while restoring the levels of GSH and total thiols with elevations in" glutathione S-transferase and superoxide dismutase. Our results indicate that CoCl2-induced erythropoiesis was accompanied by altered antioxidant status of the erythrocytes. Kolaviron, however, ameliorated the disturbancesin erythrocyte antioxidant defense system. 1 results 1

- Lead acetate (PbA) is one of the major environmental contaminants with grave toxicological consequences both in the developing and developed countries. The liver and erythrocyte antioxidant status and markers of oxidative were assessed. Exposure of rats to PbA led to significant decline (p < 0.05) in hepatic and erythrocyte glutathione peroxidase (GPx), glutathione S-transferase (GST), catalase (CAT), superoxide dismutase (SOD), and reduced glutathione (GSH) content. Similarly, malondialdehyde (MDA) and H2O2 concentrations were significantly (p < 0.05) elevated. Histopathology and immunohistology of liver of rats exposed to PbA showed focal areas of necrosis and COX-2 expression after 6 weeks of PbA withdrawal. Taken together, hepatic and erythrocytes antioxidant defence system failed to recover after withdrawal of the exposed PbA for the period of the study. In conclusion, experimental animals exposed to PbA did not recover from hepatotoxicity and disruption of erythrocyte antioxidant defence system via free radical generation and oxidative stress. 1 results 1

- The harmful effects of folkloric uses of Nigerian bonny light crude oil (BLCO) in ailments management may outweigh the expected beneficial effects. We investigated the levels of heavy metal concentrations in the tissues as well as the effect of BLCO on the antioxidant status of erythrocytes of rats after oral exposure to 0, 200 and 800 mg/kg BLCO for 7 days. Analysis of heavy metal concentrations in BLCO showed that Zn > Fe > Pb > Cu > Ni. The trend of accumulation of the metals in the tissues is blood—Fe > Pb >Zn whereas Cu and Ni levels were not affected; Liver—Ni > Zn > Fe > Cu > Pb and Testes—Ni > Cu > Pb > Zn > Fe. The order of concentration of the metals in the tissues is as follows: iron—blood > liver > testes; zinc—liver > blood > testes; lead—blood > liver > testes; copper—testes > liver > blood; nickel—liver > testes > blood. Activities of the antioxidant enzymes of erythrocytes such as superoxide dismutase, catalase, glutathione S-transferase and glutathione peroxidase increased significantly in a dose-dependent manner with significant elevation in hydrogen peroxide and malondialdehyde levels, whereas glutathione level was not significantly decreased in BLCO-treated animals. Collectively, the results showed that BLCO induces oxidative damage to erythrocytes of rats. 1 results 1
- This study evaluated the ameliorative potential of aqueous extract of Spondias mombin against arsenic-induced toxicity in the rat brain. Forty-five albino rats were randomly divided into nine groups of 5 rats each. Groups A, C and E were administered S. mombin leaf extract alone in graded doses of 100, 200 and 300 mg/kg body weight respectively for 7 days. Groups B, D and F were administered the extract at 100, 200 and 300 mg/kg respectively for 7 consecutive days in a single oral dose by gavaging before oral administration of sodium arsenite (NaAs02) (2.5mg/kg) on day 7. Groups G and H served as the negative control groups and received 0.2 ml diluted propylene glycol (vehicle for the extract) and distilled water respectively. Group I received distilled water for 7 consecutive days and 2.5mg/kg NaAs02 as a single oral dose on the 7th day. Heamatological (packed cell volume, red blood cell count, haemoglobin, white blood cell count, platelets, and neutrophils count) and biochemical parameters (serum alkaline phosphatase, aspartate aminotransferase, alkaline phosphatase, gamma glutamyl transferase) urea and Creatinine were evaluated in all animals. Clastogenecity activity was evaluated by studying micronuclei formation in polychromatic erythrocyte cells in bone marrow. Pretreatment with S. mombin significantly reduced the elevated serum levels of liver enzymes and reduced the frequency of micronucleated polychromatic erythrocytes (MnPCEs) in rat bone marrow intoxicated with arsenic. Histological examinations showed that the extract at tested dosages protected against NaAs02-induced liver damage. Our findings suggest that the leaf extract of S. mombin possesses a remarkable ameliorative effect against sodium arsenite induced toxicity in albino rats. 1 results 1
