The major findings are: (1) The identification of a peptide GnRH antagonist with differential affinity between human and sheep GnRH receptors, which could be used in future studies to map its binding site. (2) The identification of a set of residues which contribute to the differential affinity of the non-peptide GnRH antagonist FE 101177 towards sheep and human receptors. (3) The synthesis and chracterization of PAnt-1, a novel photoreactive peptide GnRH antagonist with high affinity and cross/inking efficiency (4) The localization of the attachment site of PAnt-1 to a 7-residue segment of the Nterrninal domain of the receptor, with Cys14 the putative cross/inking site.…”
