ENGLISH ABSTRACT: This body of work entails the synthesis and characterisation of non-alternating poly(styrene-co-maleic anhydride) copolymers via an iterative RAFT technique. This approach affords copolymers which are characterised by a narrow chemical composition distribution. After basic hydrolysis, the synthesised copolymers were then employed in lipid solubilization studies in order to evaluate their efficiency in styrene-maleic acid lipid particle (SMALP) formation. Relative to commercially available poly(styrene-co-maleic anhydride), the RAFT-made polymers yielded SMALPs with improved thermal stabilities and were found to insert more efficiently into lipid monolayers. The improved solubilisation efficiency of the RAFT-made polymers was ascribed to the homogenous distribution of monomer units throughout the polymer backbone. This arrangement is said to promote uncoiling of the styrene-rich hydrophobic core allowing access to the specific conformation required for nanodisc stabilisation. AFRIKAANSE OPSOMMING: Hierdie werkstuk behels die sintese en karakterisering van nie-afwisselende poli (stireen-ko-maleïne-anhidried) kopolimere via 'n iteratiewe RAFT-tegniek. Hierdie benadering bied kopolimere aan wat gekenmerk word deur 'n nou verspreiding van chemiese samestellings. Na basiese hidrolise is die gesintetiseerde kopolimere dan in lipiedoplosbaarheidstudies gebruik om hul doeltreffendheid in die vorming van stireenmaleïnezuur-lipieddeeltjies (SMALP) te evalueer. Met betrekking tot die kommersieel beskikbare poli(stireen-ko-maleïne-anhidried) het die RAFT-vervaardigde polimere SMALP's met verbeterde termiese stabiliteit opgelewer, en dit voeg meer doeltreffend in die lipiedmonolaag. Die verbeterde oplosbaarheid van die RAFT-vervaardigde polimere word toegeskryf aan die homogene verspreiding van monomeereenhede deur die polimeer ruggraat. Daar word beweer dat hierdie rangskikking die afwikkeling van die stireenryke hidrofobiese kern bevorder, waardeur toegang tot die spesifieke konstruksie benodig word vir nanodisk stabilisering.
