ENGLISH ABSTRACT: Cancer has always been a serious problem in the medical field. Certain types of cancer such as pancreatic cancer, has a very low survival rate. One of the reasons for this is because it is typically detected at a very late stage making it nearly im- possible to cure. With the increasing amount of research in the development of biosensors, it has become a possibility to use this technology for the early detec- tion of diseases like cancer which should improve the chances of survival. This project aimed to develop a biosensor for the early detection of pancre- atic cancer. Some of the main goals were to identify a suitable biomarker and transducer for further investigation. After establishing glypican-1 in circulating exosomes as the biomarker, and electrochemical analyses as the transducer, a sec- ondary goal was defined to evaluate the feasibility of using specifically orien- tated antibodies over random orientated antibodies on the biosensing surface. For random orientated antibodies, a protocol using EDC (1-ethyl-3-(3-dimethyl amino-propyl) carbodiimide) and NHS (Ny-hydroxysuccinimide) was used as investigation together with a protocol using cysteamine and glutaraldehyde. For specifically orientated antibodies, a protocol was established where TCEP (tris(2- carboxyethyl) phosphine) was used to split antibodies. It was found that using specifically orientated antibodies for biosensor devel- opment significantly improved measurements compared to randomly orientated antibodies. The final biosensor successfully detected glypican-1 to a concentra- tion of 0.1 ng/mL. The sensor also managed to differentiate glypican-1 from CA 19-9, but failed to detect different concentrations of glypican-1 and should there- fore be the main focus of future research. AFRIKAANSE OPSOMMING: Kanker was nog altyd ’n ernstige probleem in die mediese veld. Sekere tipes kan- ker, soos pankreaskanker, het ’n baie lae oorlewingsyfer. Een van die redes hier- voor is dat dit tipies baie laat opgespoor word wat dit byna onmoontlik maak om te genees. Met die toenemende hoeveelheid navorsing in die ontwikkeling van biosensors, het dit ’n moontlikheid geraak om hierdie tegnologie te gebruik om siektes soos kanker vroeg op te spoor wat die kanse van oorlewing kan verbeter. Hierdie projek was gerig daarop op om ’n biosensor te ontwikkel om pankre- askanker op ’n vroeë stadium op te spoor. Sommige van die doelwitte was om ’n gepaste biomarker en omskakelaar te identifiseer vir verdere ondersoek. Nadat dit vasgestel is dat glypican-1 in sirkulerende eksosome ’n gepaste biomerker is, en elektrochemiese analises ’n gepaste omskakelaar is, was ’n sekondêre doelwit vasgestel om uit te vind of dit meer lewensvatbaar is om spesifiek geörienteerde teenliggaampies te gebruik op die biosensor oppervlak in vergelyking met luk- raak geörienteerde teenliggampies. Vir lukraak geörienteerde teenliggaampies, was ’n protokol wat EDC en NHS insluit, gebruik tesame met ’n protokol wat sisteamien en gluteraldehied gebruik. Vir geörienteerde teenliggaampies was ’n protokol vasgestel wat TCEP gebruik om teenliggaampies te verdeel. Daar is gevind dat spesifiek geörienteerde teenliggaampies die metings van die biosensor aansienlik verbeter het in vergelyking met lukraak geörienteerde teenliggaampies. Die finale biosensor kan glypican-1 suksesvol identifiseer tot en met ’n konsentrasie van 0.1 ng/mL. Die sensor kan ook differensieer tussen glypican-1 en CA 19-9. Die sensor het wel misluk daarin om verskillende kon- sentrasies van glypican-1 te onderskei wat dus die hooffokus moet wees van toe- komstige navorsing.
