Several experimental studies now exist indicating that oxygen radicals contribute to Mycardial damage in ischaemia and reperfusion. The majority of these studies provided indirect evidence of oxygen radical involvement by using radical scavengers or specific enzymes (e.g. catalase, superoxide dismutase). Recent studies, using electron spin resonance, provided direct evidence of a "burst" of free radical generation especially during reperfusion. Whether generation of free radicals during ischaemia alone is solely responsible for the membrane changes characteristic of myocardial ischaemia, remains to be established. The aim of this study was therefore to compare the well-known ischaemia-induced membrane changes with those produced by an in vitro free radical attack on either isolated myocardial membranes or the perfused rat heart.
