The anaesthetic, propofol and the opioid, alfentanil have pharmacokinetic profiles suitable for total intravenous anaesthesia. The hypothesis states that they are cardiovascular depressants and that their administration causes haemodynamic instability. A review of the literature is presented wherein the pharmacokinetics and the cardiovascular pharmacodynarnics of the drugs are amplified. Pig pharmacokinetic models for propofol and alfentanil, were determined by applying curve-fitting computer programs to measured post-infusion concentrations and were used by computer-controlled infusions to establish predicted drug concentrations during haemodynamic studies.
