Atrial natriuretic peptide (ANP), a potent natriuretic, diuretic and vasorelaxant factor, is secreted from the mammalian heart. Atrial secretory granules (ASG), the intracellular storage sites for ANP, are a vital component of the cardiac endocrine system. Regulated secretion of ANP involves controlled release of the peptide from this storage site in response to stimulation. Mechanisms regulating this critical step in the ANP secretory pathway are poorly understood. In other endocrine organs, such as the adrenal medulla, calcium plays an important role in the control of the secretory pathway and this function is mediated by calcium-binding proteins. However, in the cardiac endocrine system the role of calcium is controversial. The main focus of this thesis was the identification and characterization of calcium-binding proteins that may regulate the calcium dependent secretion of atrial natriuretic peptide (ANP) by interaction with atrial secretory granules. Due to the controversy surrounding the role of calcium in this secretory process, the calcium dependence of ANP secretion was first addressed before proceeding with the isolation and characterization of calcium-dependent atrial granule-binding proteins from rat hearts.
