Neuroendocrine abnormalities associated with depression include elevated circulating cortisol levels and non-suppression of cortisol secretion following the administration of dexamethasone. These aberrations in cortisol secretion suggested that the hypothalamic-pituitary-adrenal (HPA) axis may be hyperactive in depressed patients. In view of this proposal, the present study was undertaken in order to investigate possible mechanisms in the central nervous system which could alter the activity of the HPA axis. Since the central noradrenergic system has been implicated in the etiology of depression, a noradrenaline re-uptake- blocker, as well as noradrenergic agonists and antagonists, were administered to rats and their effects on corticosterone secretion were determined. lntraperitoneal (i.p.) injection of either desimipramine (20 mg/kg), a noradrenaline re-uptake blocker, or yohimbine (3 mg/kg, i.p.), an a2-adronoceptor antagonist, resulted in the stimulation of corticosterone secretion 15, 30 and 60 min after injection of the drugs. Th eplasma corticosterone concentration of rats that had received combined treatment with desimipramine and yohimbine, was not only significantly greater than the plasma corticosterone concentration of their saline treated controls at all time intervals, but was also significantly greater than the 60 min plasma corticosterone value of those rats that had received i.p. injections of disimpramine or yohimbine alone. The i.p. injection of isoproterenol (1 mg/kg), a B-adrenoceptor agonist, significantly elevated the plasma corticosterone concentration, which was prevented by the prior administration of propranolol (10 mg/kg), a B-adrenoceptor antagonist. Both the i.p. injection and the direct injection of clonidinc into the paraventricular nucleus of the hypothalamus (PVN) increased corticosterone secretion dose-dependently. Yohimbine (1 mg/kg) given prior to clonidine (250ug/kg, i.p.) inhibited the clonidine-induced stimulation of corticosterone secretion.
