Department of Biomedical Sciences: Molecular Biology and Human Genetics Chromatin Dissertations -- Medicine UCTD Thesis (PhD)--University of Stellenbosch, 1993. ENGLISH ABSTRACT: Oxygen-derived free radicals are implicated in many human diseases including cancer. The mutagenic and carcinogenic properties of these species manifest themselves in lingering damage to several biomolecules including DNA. Here, efforts have been made to chemically characterize DNA base damage and DNA-protein cross-links produced in isolated chromatin and in cultured mammalian cells exposed to ionizing radiation or to H2O2/metal ions. Gas chromatography/mass spectrometry with selected ion-monitoring was used for this purpose.
