A review is presented in chapter 1 dealing with the nuclear and morphological changes during spermatogenesis and sperm transport with emphasis on the transformation of spermatids during spermiogenesis, maturation of spermatozoa, the acrosome reaction and fertilization. A literature survey is presented of the fluorescent-binding agents Acridine Orange and Quinacrine Mustard and of the zona free hamster ovum penetration technique, which allows actual visualization of individual human sperm chromosomes. Routine semen analysis is described with special emphasis on the evaluation of sperm morphology according to stricter criteria. Two different methods determining sperm motility (visually and computerized) were described/discussed. The procedures for evaluation of sperm nuclear status, as measured by the Acridine Orange test, are also described. The technique to obtain human sperm chromosomes by the fusion of zona free hamster oocytes is also presented in this chapter.
