Serum amyloid A protein (SAA) was discovered and characterized as a major acute phase protein produced in the liver upon inflammatory cytokine stimulation. As an apolipoprotein of high density lipoprorein (HDL) it displaces apo-A1 and remodels the HDL particle during inflammation. In this thesis new members of the SAA superfamily are identified and characterized in man and mouse. This was achieved by correlating analytical protein data with the known disposition of the genes of these molecules. The SAA superfamily is shown to consist as 2 distinct subfamilies. The one subfamily encompasses the classical acute phase SAA's which are referred to as A-SAAs. The other subfamily comprises the major form of SAA on normal HDL. It is constitutively expressed and is present as minor apolipoprotein components of normal HDL. These molecules, C-SAA in man and SAA5 in the mouse, is postulated to be involved in the normal metabolism of HDL. Further characterization of the inflammatory A-SAA's was pursued in the mouse to identify an amyloid-resistant mouse strain. To this end it is shown that in the CE/J mouse which is amyloid-resistant, only a single inflammatory A-SAA molecule exists which shares characteristics of both SAA1 and SAA2. The cloning of the gene of this molecule is described and it offers the potential for site-directed mutagenesis to identify a structural motif that would allow for selective deposition of certain A-SAA's into amyloid. The studies presented here promote the understanding of the SAA superfamily which, being so well-conserved through evolution, is likely to be of biological importance.
