TEM studies of the sperm ultrastructure of three South African echinoderms (Parechinus angulosus (Leske, 1778): Echinoidea; Ophiarachnella capensis (Bell, 1888): Ophiuroidea; Patiriella exigua (Lamarck, 1816): Asteroidea) were performed. All three species have a sperm morphology of the primitive type consisting of spherical (O. capensis and P. exigua) or conical (P. angulosus) headpieces, a midpiece and a long flagellated tail. The shape of the apical granules varies from the semi-spherical for P. angulosus, to flattened ovale for O. capensis to spherical for P. exigua. These are enclosed in an acrosomal membrane and are respectively embedded in cup shaped, undulating and funnel shaped nuclear fossa. The posterior contingent of the peri-acrosomal material of the echinoid and asteroid sperm is differentiated as a sub-acrosomal component, while it appears homogeneous in the ophiuroid sperm. Nuclear vacuoles are found in the electron dense and granular nuclei of the non-echinoid sperm, but seem to be absent from Parechinus sperm. The latter contain a deep posterior nuclear fossa that is less prominent in Ophiarachnella sperm and absent from Patiriella sperm. A single, annular mitochondrion encircles the sperm flagellum of P. angulosus and O. capensis symmetrically and that of P. exigua asymmetrically. Two centrioles (distal and proximal) are associated with the flagella of each of the three species. Using suitable micrographs, the typical 9+2 configurations of microtubuli and accessory structures of flagella are described. The angle at which the flagellum of P. angulosus and O. capensis joins the headpiece is perpendicular to the nuclear basis and less than 90° in the P. exigua sperm. In all three species the sperm cell is totally enclosed by a plasma membrane except where it fuses with the apical membrane to form the compound membrane. In order to show phylogenetic and taxonomic similarities and differences, sperm ultrastructure was compared amongst the three species and with other species of the same classes. In the second part of this study, the influence of lindane on the sperm ultrastructure of P. angulosus was investigated. Appreciable damage to the membrane and nuclear material of the sperm of study animals, exposed to 500 µg/l lindane, was observed and described. These findings support evidence that sperm morphology could possibly be used as a biomarker of environmental quality.
