This study was undertaken to investigate the possible presence of an aziridine in S2, a biological active fraction first isolated by Swart from the plant Salsola tuberculatiformis Botsch. It was known that S2 contains a certain circumstances decomposes to the less active 1-(4-hydroxyphenyl)-2-(methylamino) ethanol, also known as synephrine. The strategy followed to prove the presence of the aziridine was based on the use of an aziridine probe, containing a reactive functional group that reacts with aziridines but which leaves synephrine unaffected. Six different probes were designed and synthesized for this purpose. Two model aziridines, 2-phenyl-aziridine and 1-methyl-2lphenyl-aziridine were also prepared and tested with each of the probes. The results showed that out of the six, the aziridine probe N-(2,4,6-trinitrophenyl)-mercaptoethanol-amine (N-TNP-MEA-SH) complied best with the requirements. The probe N-TNP-MEA-SH was then used to investigate the S2 fraction. The results clearly indicated that S2 contains a component reacting with the aziridine probe. This proves in a chemical way that S2 contains a compound which differs from synephrine. The product arising out of the reaction between S2 and N-TNP-MEA-SH was chemically unstable. During isolation on preparative thin layer plates, it rapidly decomposed to yield the original probe plus a new compound which could be detected on the thin layer plate. The latter product was identified using mass-spectroscopy as the unsaturated compound, 2-(4-hydroxyphenyl)-1-(aminomethyl)ethene. All the findings of the present study are in accord with the presence of 2-(4-hydroxyphenyl)-1-methylaziridine, or a precursor compound which converts to this aziridine, in the fraction S2. The successful identification of the aziridine in S2 places the investigation of the nature and biological effects of the active natural products in Salsola tuberculatiformis Botsch. on a much firmer footing. The aziridine probe developed in this study will in future be used in an extended search for aziridine containing natural products in Salsola and in other plants. This study, mo doubt, will also lead to the synthesis of 2-(4-hydroxyphenyl)-1-methylaziridine and precursors of this compound.
